What actually happens
Two brothers and a long drive
Seth Gecko and his unstable brother Richie are bank robbers on the run. At a liquor store in Texas, they shoot the clerk and a Texas Ranger and wreck the building, then drive to a motel with a hostage, a bank teller. When Seth goes out for dinner, Richie kills her. Jacob Fuller, a widowed pastor who doubts his faith, checks into the same motel with his teenage kids Scott and Kate. Seth sizes up their motor home, kidnaps the family and orders them to drive to Mexico. They have a tense encounter at the border, and the guard lets them through. They stop at a desert strip club called the Titty Twister, where the brothers' contact Carlos is due to collect them in the morning and take them to a hideout town called El Rey. The doorman tries to stop them, and the brothers overpower him.

The part where the movie changes genres
Inside the club are a biker named Sex Machine, who keeps a pistol in his pants, and Frost, a Vietnam veteran. The bartender says only bikers and truckers can drink there, and Jacob argues that his motor home makes him a trucker. The bartender accepts this. They sit down for a performance by the headliner, Santánico Pandemonium. Once it ends, the doorman orders the group out, Richie goes for someone, and the bartender stabs him in his injured hand. Seth shoots the bouncer, and then the doorman, who laughs at the brothers first. This is the moment people dread, and it happens right after the fight breaks out: Santánico sees Richie's bloody hand and turns into a monstrous vampire, then jumps on him.
Everyone gets a wooden stake
The other dancers turn out to be vampires too, and they start feeding on the customers while the dead staff get up again. Kate kills the doorman with her cross pendant, and Seth kills Santánico by shooting a wooden chandelier down onto her. Sex Machine and Frost stab several vampires with bits of wood. When the room is clear, the survivors find the door blocked and bats swarming outside. Richie rises from the dead, and Seth tries to protect him until Jacob says there is no saving him, and Seth drives a stake into his heart. Later, while Frost tells a war story, Sex Machine, who was bitten earlier, turns and bites Frost and then Jacob. Frost throws him through a high window, which lets the bats in, and Frost then turns too. Seth, Kate and Scott retreat to a storeroom.
Sunrise, and Carlos finally shows up
Jacob covers their retreat with a shotgun shaped like a cross and joins them in the storeroom. They build weapons from the cargo of trucks that earlier victims left behind. Jacob knows he will turn soon, so he gets Scott and Kate to swear they will put him down when it happens. The group then fights the vampires one more time. Sex Machine becomes a rat-like monster and attacks Seth, and Kate kills him. Jacob kills Frost and then turns, and Scott kills Jacob but hesitates just long enough to be bitten. The other vampires get Scott, and Scott begs for death, so Kate shoots him. When Kate and Seth are surrounded, morning light comes through holes in the walls and makes the vampires retreat. Then Carlos shows up, and his bodyguards open the door to let in full sunlight.
The ending, explained
The sunlight kills the remaining vampires. Seth complains that Carlos picked a terrible meeting place, haggles over a lower fee to get into El Rey, and pays. He hands Kate money and says sorry. She offers to go along with him, and he turns her down, saying he is not that immoral. Afterward Kate leaves alone in the motor home. As she goes, the camera reveals that the Titty Twister is really the peak of a buried Aztec temple, with abandoned trucks from several decades parked around it. Richie, Jacob, Scott, Frost, Sex Machine and Santánico are all dead.
